    New Discovery Shows Human Cells Can Write RNA Sequences Into DNA – Challenges Central Principle in Biology

    A recent discovery reveals that human cells can transcribe RNA sequences into DNA, challenging a central principle in biology.

    In a discovery that challenges long-held dogma in biology, researchers show that mammalian cells can convert RNA sequences back into DNA, a feat more common in viruses than eukaryotic cells.

    Cells contain machinery that duplicates DNA into a new set that goes into a newly formed cell. That same class of machines, called polymerases, also build RNA messages, which are like notes copied from the central DNA repository of recipes, so they can be read more efficiently into proteins. But polymerases were thought to only work in one direction DNA into DNA or RNA. This prevents RNA messages from being rewritten back into the master recipe book of genomic DNA. Now, Thomas Jefferson University researchers provide the first evidence that RNA segments can be written back into DNA, which potentially challenges the central dogma in biology and could have wide implications affecting many fields of biology.

    “This work opens the door to many other studies that will help us understand the significance of having a mechanism for converting RNA messages into DNA in our own cells,” says Richard Pomerantz, PhD, associate professor of biochemistry and molecular biology at Thomas Jefferson University. “The reality that a human polymerase can do this with high efficiency, raises many questions.” For example, this finding suggests that RNA messages can be used as templates for repairing or re-writing genomic DNA.

    The work was published June 11th, 2021, in the journal Science Advances.

    A Focus on the Unusual Polymerase Theta

    Together with first author Gurushankar Chandramouly and other collaborators, Dr. Pomerantz’s team started by investigating one very unusual polymerase, called polymerase theta. Of the 14 DNA polymerases in mammalian cells, only three do the bulk of the work of duplicating the entire genome to prepare for cell division. The remaining 11 are mostly involved in detecting and making repairs when there’s a break or error in the DNA strands. Polymerase theta repairs DNA, but is very error-prone and makes many errors or mutations. The researchers therefore noticed that some of polymerase theta’s “bad” qualities were ones it shared with another cellular machine, albeit one more common in viruses — the reverse transcriptase. Like Pol theta, HIV reverse transcriptase acts as a DNA polymerase, but can also bind RNA and read RNA back into a DNA strand.

    In a series of elegant experiments, the researchers tested polymerase theta against the reverse transcriptase from HIV, which is one of the best studied of its kind. They showed that polymerase theta was capable of converting RNA messages into DNA, which it did as well as HIV reverse transcriptase, and that it actually did a better job than when duplicating DNA to DNA. Polymerase theta was more efficient and introduced fewer errors when using an RNA template to write new DNA messages, than when duplicating DNA into DNA, suggesting that this function could be its primary purpose in the cell.

    Structural Flexibility Supports RNA Binding

    The group collaborated with Dr. Xiaojiang S. Chen’s lab at USC and used x-ray crystallography to define the structure and found that this molecule was able to change shape in order to accommodate the more bulky RNA molecule — a feat unique among polymerases.

    “Our research suggests that polymerase theta’s main function is to act as a reverse transcriptase,” says Dr. Pomerantz. “In healthy cells, the purpose of this molecule may be toward RNA-mediated DNA repair. In unhealthy cells, such as cancer cells, polymerase theta is highly expressed and promotes cancer cell growth and drug resistance. It will be exciting to further understand how polymerase theta’s activity on RNA contributes to DNA repair and cancer-cell proliferation.”

      In order for RNA molecules (like viral RNA or RNA bases vaccines) to change the nuclear DNA, several conditions must be met: it must be reverse transcribed (which this paper shows may be possible), it must be integrated into the DNA (which this paper does not show), and it must be localized to the nucleus. So before everyone gets up in arms claiming that the mRNA COVID vaccines can in fact change our DNA, realize that this paper only shows that one of the three conditions required to do is demonstrated by these researchers.

        If something is pushed in a hurry with a political agenda (certainly is not science), do the smell test.

        In 1918 most people died of bacterial pneumonia, not from the Spanish Flu of 1918 (debilitating factor).

        In 1976 and the rush to vaccinate, which had to be stopped because once again the vaccine was killing more people than the disease.

        Eugenics and dysgenics are the name of the game.

              They’re argument is akin to jumping off a cliff. The ground is the so-called “killer” (bacterial pneumonia) but I don’t think a reasonable person would advise the jump. Get rid of the jump (Spanish Flu) the ground is innocuous.

        I have seen a few people ask this. No it does not. The article only states that the enzyme studied appears in large quantities in drug resistant cancer cells. So the take away here is that we have a potential new avenue to fight cancer drug resistance following more research done with this enzyme.

      HOW DOES DNA TURN A CELL INTO A SHEEP, OR A BIRD, OR A HUMAN?

      When you divide a cake, the cake never gets bigger. However, when we were just a single cell and that cell kept dividing we got bigger. New material had to come from somewhere. That new material came from food.

      Just as the sequence of various letters and words in human language communicate a message and direct workers to build and assemble something so, too, the sequence of various molecules in our DNA (our genes or genetic code) directed the molecules from our mother’s food, that we received in the womb, to become new cells, eventually forming all the tissues and organs of our body.

      When you feed a cat your food the cat’s DNA will direct the food molecules to become the cells, tissues, and organs of a cat, but your DNA will turn the same food into human cells, tissues, and organs.

      This article will give you a good understanding of how DNA, as well as cloning and genetic engineering. You also learn that so-called “Junk DNA” isn’t junk at all. You will learn why it is not rational to believe that DNA code could have arisen by chance. Science points (not proves, but points) to an intelligent cause for DNA code.

      What about genetic and biological similarities between species? Genetic information, like other forms of information, cannot happen by chance, so it is more logical to believe that genetic and biological similarities between all forms of life are due to a common Designer who designed similar functions for similar purposes. It doesn’t mean all forms of life are biologically related! Only genetic similarities within a natural species proves relationship because it’s only within a natural species that members can interbreed and reproduce.

      Nature cannot build DNA code from scratch. It requires already existing DNA code to direct and bring about more DNA code or a genetic engineer in the laboratory using intelligent design and highly sophisticated technology to bring DNA code into existence from scratch. Furthermore, RNA/DNA and proteins are mutually dependent (one cannot come into existence without the other two) and cannot “survive” or function outside of a complete and living cell. DNA code owes its existence to the first Genetic Engineer – God!

      Protein molecules require that various amino acids come together in a precise sequence, just like the letters in a sentence. If they’re not in the right sequence the protein won’t function. DNA and RNA require for various their various nucleic acids to be in the right sequence.

      Furthermore, there are left-handed and right-handed amino acids and there are left-handed and right-handed nucleic acids. Protein molecules require for all their amino acids to be left-handed only and in the right sequence. DNA and RNA require for all their nucleic acids to be right-handed and in the right sequence. It would take a miracle for DNA, RNA, and proteins to arise by chance!

      Mathematicians have said any event in the universe with odds of 10 to 50th power or greater is impossible! The probability of just an average size protein molecule (with its amino acids in the right sequence) arising by chance is 10 to the 65th power. Even the simplest cell is made up of many millions of various protein molecules along with and DNA/RNA..

      The late great British scientist Sir Frederick Hoyle calculated that the odds of even the simplest cell coming into existence by chance is 10 to the 40,000th power! How large is this? Consider that the total number of atoms in our universe is 10 to the 82nd power.

      Also, so-called “Junk DNA” isn’t junk. Although these “non-coding” segments of DNA don’t code for proteins, they have recently been found to be vital in regulating gene expression (i.e. when, where, and how genes are expressed, so they’re not “junk”). Also, there is evidence that, in certain situations, they can code for proteins through the cell’s use of a complex “read-through” mechanism.

      Reverse transcriptase has been known for about a decade. HIV-1 has been found to have it’s RNA transcribed into the cells genome. I love how this has completely been ignored as soon as all the world (except China who used classic deactivated virus) developed RNA and mRNA vaccines based off S-spike protein. If this via reverse transcriptase was incorporated into the cellular genome, the potential for the cells to continually produce S-spike proteins which have a unique affinity for ACE-2 receptors which are primarily located in the vascular system used to regulate blood pressure amongst many other functions it could pose serious danger. Recent autopsy of fully vaccinated patient found their body was completely overloaded with the S-spike protein which caused organ and cardiovascular failure. Studies have shown by using a dead virus with no RNA only a S-spike coated shell that it caused the same damage that Covid-19 did on various cell-lines I believe they used lung cell lines. This must be fully investigated and no ignored because this may throw the entire reasoning behind this recent vaccination development into question.

        You claim what is NOT claimed by the study you refer to. The study found the patient was infected with SARS-CoV-2 by means of the positive RT-PCT testing for various tissues. It does not show that the patient was overloaded with spike protein.

        Indeed, what the other reply said: the patient was actually infected with the virus, “RNA was extracted from paraffin sections using the Maxwell RSC (Promega, Madison, WI, USA). Multiplex RT-PCR analysis targeted 2 independent genes of the SARS-CoV-2-genome (Fluorotype SARS-CoV-2 plus Kit; HAIN/Bruker, Nehren, Germany): RNA-dependent RNA polymerase (Target 1) and nucleopeptide (Target 2).”, they tested for 2 different SARS-CoV-2 genes and the tests came back positive for both. The patient was an 86 year old man with multiple pre-existing health conditions.

        What the paper suggests is that immunogenicity is not enough to protect from the virus and that sterile immunity is required: i.e. especially if you are an older individual and have reduced immune capacity, your body needs to have finished mopping up the spike proteins produced in response to vaccination, in order to have the antibody capacity to bind and eliminate the actual virus. This is of course an oversimplification, but you can imagine that when the body is fighting two wars that both start around the same time, it’s less effective in both.

        The article you linked only shows that after a single dose of vaccin you can get infected by the coronavirus (not by the vaccin) and develop the well-known disease. What they found in the body is the virus, not the vaccin. This man got the covid after having a first dose of Pfizer vaccin. That’s it.

      All this takes place during cell division as stated in the article. mRNA does not enter the nucleus. It only instructs the cell to make a single protein and nothing else. Afterwards the mRNA strand gets destroyed by the cell.

      The last paragraph in this article is worrying. What is NIH upto? I think we need some clarity that this tech is not what is in the recent jabs. If that isn’t the case, then what changes are being made to human DNA and if DNA to DNA replication is flawed, what errors are being produced.

      This is revolutionary..doors are open to again think the central dogma. Now can hypothesize that the genome keeps on changing after several cycles of division and replication..the introns or untranslated RNAs might have some specific sequences or markers that might again correct the entire specific gene after the above mentioned replication and division cycle..with the help of polymerase theta this work can be done.
      This is one kind of more accuracy to keep the DNA unchanged.

      Does this not support the theory that viruses themselves are actually manufactured in our bodies in response to various environmental factors?

      If the body can make strands of RNA, then how do we know the RNA strands that we call virus are not manufactured in our bodies as well.

      The evidence that viruses originate anywhere besides living cells is extremely lacking to say the least.

      Perhaps Bechamp was right, and Pasteur wrong after all.

      This would explain how the flu seems to only occur during the winter except in rare cases, along with many other things about virus that do not hold up to intense scrutiny.

      Despite our societies nearly 100% belief that foreign particles from outside our body cause most diseases there is NOT a single replicated experiment that supports this theory.

      Just read about Pasteur trying to make the “polio vaccine” and how much trouble he had trying to transmit the disease.

      Nothing worked until they drilled holes in the animals skull and injected what was basically the mucous of an animal with polio into the hole.

      Leave it to the tinfoil hat antivaxx brigade to chime in with the failed biology class comments. The mRNA is translated in the cytosol, so there is no need for the RNA to enter the cell nucleus, and the risk of being integrated into the host genome is averted. Regardless of this, the pandemic will continue until more people are vaccinated along with a certain level of herd immunity. I’ve had several healthy people in my life either die or are still suffering from the after effects from Covid-19. If you still believe that it’s a political hoax, you are living in a fantasy world that will eventually come crashing down.

        I rarely leave comments, BUT you can say all is well with you and insult others, BUT my uncle has his MD, spent 25 years as a drug researcher for GlaxoSmithKline. He says, don’t get the vaccine. I am not saying I do or do not believe in this issue being a problem for the vaccine, but I am pretty effing good at Biology, and History. So 1. The article says that this discovery does what… OH! Challenges a Central Principle in Biology. The fact that any of you thinks that can’t happen again(!) Is embarrassing. 2. A little History fun, about that government regulated H1N1 vaccine effort of 1976 where, yes, more people died from the vaccine than the actual flu. Interestingly, an article by the CDC says one takeaway is that there should only be health officials, not politics involved…
        Sadly so many don’t know enough either way. So, mind yours, because no, not enough is known for some.

      I do see comments from people expressing concerns on the possibility of theta polymerase coding the mRNA into DNA. If such a worry has any substance, then the same would be applicable during the real infection of the RNA virus too. Isn’t it?

      This has nothing to do with you refusing the COVID vaccine. Don’t try to do science with no knowledge or experience in scientific thinking whatsoever. LETS assume that the mRNA vaccine changes ur DNA (it doesn’t, mRNA is a middle phase that just vanishes after use) it would effect a really small part of your somatic (body) cells that does not represent your genetic make-up at all. Mutations happen all the time, almost all of them just fizzle out, so that’s another NBD with an “if”.

      Don’t fki try to do science with facebook articles, then call people sheeple.

      The jabs are recombinant ( recombined DNA). They are wrapping it in nano particles little computers and also have aborted fetal cells in them. The DNA is now know to be like binary code ( computer code) so they can code the nano particles or program them to cut the person’s DNA and insert the new DNA strand…what DNA ?? Who’s DNA ?? Who knows ?? They use crisper( cas9 ) gene editing as well as it is a cheap gene editing which they have been using for several years now. The jabs are loaded with LuciferASE which illuminates and helps track how the process is working when put under florescent lights ..pretty much your insides light up like a lightening bug butt. They put silver which makes it attach and seals the DNA back together. I can’t see anything good about cutting a person DNA . I don’t know how it will affect our future but it don’t look good.

      I agree with the person who said the comments here are embarrassing. It’s unfortunately the usual distrust of science, selective paranoia and lack of perspective on the risks of life. I have preconditions and I jumped at the opportunity for the vaccine. I’ve felt fine for over 4 months! If I die of either, well it’s the end of the road. I don’t want to die stupid and anti-everything that is proven to help. I already probably ingested things my whole life that harmed my DNA. So many millions of people would take vaccines in hard-hit countries. So they can have yours.

      I’m not a scientific expert but the cases are dropping nicely since the vax. The vax wasn’t developed by Walmart employees. My family and friends have taken the vax. We are not paranoid or anti-govt eventhough we don’t agree with some political business, we believe in being supportive citizens. Random massive population control? Oh the ignorance of the peasants. Workers and taxes are what feed the machines. mRNA. And ppl eat food at restaurants where they have no clue what greasy bums handled their food or what’s in it.

      for those curious about the mRNA vaccines, you all have mRNA in your cells in order for your body to work and these don’t integrated into the genome.

      It’s nice to see everyone so interested in science and please continue to educate yourselves, I would only advise to start learning the basics and then moove forward. From what I’ve read here not many people know the composition of a cell, it’s internal mechanism or functions or indeed how to differentiate between them. Let’s be honest if you where presented with a schematic of a plant cell and one of an animal cell would you be able to tell which is which? Or explain in detail what the parts are and what they’re doing? Talking about things you do not fully understand it’s like trying to change a bolt on a train with a bolt from an airplane then wandering if it will make the train fly. Before anyone asks I do have a background in genetics but I can also tell you that I survived Chernobyl, I glow in the dark, literally; what do you think a silly vaccine can do to me? Anyway have a great time guys and enjoy the wonders of the science!

      So, does this mean DNA could be reprogrammed to change physical attributes? Maybe change sex or bring about artificial immunities once better understood, in other words, to artificially evolve? I don’t entirely understand the biology behind it but I know DNA effects our biological makeup in big ways, tells our body how to grow and behave, so the next step for me is biological augmentation (after significant testing, of course).

      This isn’t about conspiracy theories….it’s about technological capability. Are the current vaccines designed to convert mRna into DNA? Probably not….but that’s not the issue. The issue is that we know whether or not mRna technology has the ABILITY to introduce the enzyme through reverse transcription to do so. This study does more than just raise the question. As with most technological and scientific break-throughs, there are countless examples of what good can come from the break-through….but there is also the unintended consequences to consider. We’re talking about having the ability to literally alter a person’s biological make-up, and the masses are attempting to shout down anyone who dares ask a question.

      Very few RNA viruses are double-strand RNA viruses….Coronavirus is not a double-strand virus, what is called a “retrovirus”. Therefore, getting COVID naturally cannot alter the DNA. The discussion here is about a vaccine, through reverse transpriptase, having the ability to introduce a specific enzyme that CAN write specific RNA sequences into DNA. That ABILITY is real. That ability/technology can also be used to target many other disease states, not just sars cov 2, and it can also be used to trigger SPECIFIC protein production at the cellular level. The application goes far, far beyond stopping this pandemic.
